Genome Stability: A Self-Sufficient DNA Repair Machine

چکیده

The repair of DNA double-strand breaks often requires the broken ends to be processed prior to religation. New results describe a bacterial enzyme with processing and rejoining activities encoded in a single polypeptide chain.

Von Neumann revisited: A turing machine with self-repair and self-reproduction properties

The growth and the operation of all living beings are directed through the interpretation, in each of their cells, of a chemical program, the DNA. This program, called genome, is the blueprint of the organism and consists of a sequence of tour discrete characters: A, C, G, and T. This process is the source of inspiration for the Embryonics (embryological electronics) project, whose final object...

Kinetic Theory Approach to Modeling of Cellular Repair Mechanisms under Genome Stress

Under acute perturbations from outer environment, a normal cell can trigger cellular self-defense mechanism in response to genome stress. To investigate the kinetics of cellular self-repair process at single cell level further, a model of DNA damage generating and repair is proposed under acute Ion Radiation (IR) by using mathematical framework of kinetic theory of active particles (KTAP). Firs...

Proteasome Nuclear Activity Affects Chromosome Stability by Controlling the Turnover of Mms22, a Protein Important for DNA Repair

To expand the known spectrum of genes that maintain genome stability, we screened a recently released collection of temperature sensitive (Ts) yeast mutants for a chromosome instability (CIN) phenotype. Proteasome subunit genes represented a major functional group, and subsequent analysis demonstrated an evolutionarily conserved role in CIN. Analysis of individual proteasome core and lid subuni...

HTLV-I Tax Increases Genetic Instability by Inducing DNA Double Strand Breaks during DNA Replication and Switching Repair to NHEJ

BACKGROUND Appropriate responses to damaged DNA are indispensible for preserving genome stability and preventing cancer. Tumor viruses often target DNA repair machinery to achieve transformation. The Human T-cell leukemia virus type I (HTLV-I) is the only known transforming human retrovirus and the etiological agent of Adult T-cell Leukemia (ATLL). Although HTLV-I-transformed leukemic cells hav...

Functional consequences of DNA mismatch repair missense mutations in murine models and their impact on cancer predisposition.

Mutations in MMR (DNA mismatch repair) genes underlie HNPCC (hereditary non-polyposis colon cancer) and also a significant proportion of sporadic colorectal cancers. MMR maintains genome stability and suppresses tumour formation by correcting DNA replication errors and by mediating an apoptotic response to DNA damage. Analysis of mouse lines with MMR missense mutations demonstrates that these M...
